The record for the 20 yard shuttle at the NFL combine was set by Kevin Kasper, a wide receiver for the Iowa Hawkeyes, at the 2001 NFL Combine. He set the record with a 3.73 second time.

“The name "20-yard shuttle" is derived from the total yards that athletes travel during the drill. They start in the middle of a 10 yard distance with one hand touching the ground. The athlete then pushes off their dominant leg in the opposite direction for 5 yards. After covering this distance, as quickly as possible they reverse and go 10 yards in the opposite direction. Finally, they reverse direction again, ending the drill at the starting point. The procedure is timed.” - Wikipedia
Kasper was a walk on for the Hawkeyes in 1997; by the time he walked off the field in 2001, he was the Hawkeyes’ all-time leading receptions leader for Iowa. He finished his career with 157 catches for 1,974 yards and 11 TD.
He was picked in the sixth round (190th overall) in the 2001 NFL Draft by the Denver Broncos. He had a seven year career in the NFL and won a Super Bowl with the New England Patriots in 2004. He only started nine games in the NFL but was used as a kick returner often. You can say he had a below average NFL career.
